پیشرفت در ژنوم کروموزوم گیاه

چکیده

توالی یابی نسل بعدی(NGS)،ژنوم های در حال تکامل می باشد که دیدگاه جدیدی را در سازماندهی،تکامل و عملکرد ژنوم ارائه می دهد. تعداد ژنوم های گیاهی هدف برای توالی یابی در حال افزایش است. با این حال،بدست آوردن توالی های ژنوم کامل در گونه های ژنوم زیاد دشوار می باشد، که عمدتاً به دلیل فرایندهای خواندن کوتاه مدت بوسیله پایگاه های NGS هستند که در مواجه با DNA غنی از زنجیره تکرار ناکافی می باشند و بخش بزرگی از این ژنوم ها را تشکیل می دهند. مسئله فراوانی توالی در پلی پلویئدها ترکیب شده است که در سلسله مراتب گیاهی غالب می باشد. یک روش برای غلبه بر برخی از این مشکلات،کاهش ژنوم هسته کامل برای کروموزوم های اختصاصی اش می باشد که با استفاده از رده بندی جریان ژنوم انجام می گیرد. ثابت شده است که DNAی بدست آمده در این روش برای کاربردهای بسیاری مناسب است،از جمله طرح ریزی فیزیکی مبتنی بر PCR،هیبریدیزاسیون در محل طبیعی،شکل گیری آرایش DNA، تکامل نشانگرهای DNA و ... . ارتباط رده بندی کروموزوم با NGS فرصت هایی را برای مطالعه سازماندهی ژنوم در سطح کروموزومی مجزا برای آنالیزهای تطبیقی بین گونه های مرتبط و اعتبارسازی یکپارچگی کلی ژنوم ارائه می دهد. جدا از هدف اولیه کاهش پیچیدگی الگوها،در نظر گرفتن روش مبتنی بر کروموزوم،گروه های تحقیقاتی مستقل را برای بررسی گونه های گیاهی توانا می سازد. با توجه به اینکه تعداد گونه های گیاهی برای شناسایی رده بندی کروموروم در حال افزایش است،احتمالاً ژنوم های کروموزوم برای حوزه ژنتیک گیاهی نقش مهمی دارد.

1 توالی یابی ژنوم گیاه
دهه گذشته جهش عمده ای در آگاهی ما درباره ساختار ژنوم گیاه،عملکرد و پویایی تکاملی مشاهده شده است. محرک اصلی این توسعه،روشن سازی پایگاه توالی یابی نسل بعدی(NGS) می باشد که باعث می شود به داده های بسیاری دسترسی پیدا شود.پیشرفت در بیوانفورماتیک برای آنالیز توالی DNA لازم می باشد. اولین ژنوم گیاه که به طور کامل توالی یابی شد،مربوط به گیاه آرابیدوپسیس تالینا بود، که بخاطر ژنوم کوچک آن(حدود 150mb) انتخاب شده است.
