چکیده: Atpase واکواولار H ابتدا به عنوان یک پمپ پروتون الکتروژنی شناخته شده است که
لومن(حفره) فضاهای درون و بیرون سلولی را اسیدی می کند. PH اسیدی تولیدی
شده بوسیله V-ATpase برای محدوده وسیعی از فرایندهای سلولی و همچنین فرایندهای مستقل
از اسیدی شدن مانند ترکیب غشاء و ترشح مهم است. علاوه براین نقش های
محافظتی،مطالعات اخیر به نقش V-ATpase در تنظیم مستقیم و عملکرد مسیرهای سیگنالینگ اشاره می کند. در این
بررسی،یافته های اخیر را درباره عملکردهای V-ATpase در تنظیم رشد و فزیولوژی بافت
توصیف کردیم. مقدمه ATpase واکوالار H+ یا V-ATpase یک پمپ پروتون الکتروژنی است که لومن فضاهای درون و بیرون سلولی
را اسیدی می کند. بیش از 30 سال پس از کشف اولین آنزیم از غشاءهای واکوئل مخمر می
گذرد. وجودش در مخمر نشان می دهد که آنزیم برای فیزیولوژی اولیه سلولی لازم است.
در واقع،آنزیم دارای یک نقش محافظتی می باشد. V-ATpase در ارگانیسم
های تک سلولی و چندسلولی از جمله سلولهای گیاهان و جانوران وجود دارد. این آنزیم
در محل های مختلف سلولی و بویژه در ارگانل های درون غشایی مانند لیزوزوم
ها،آندوزوم ها،دستگاه گلژی،گرانول های ترشحی و کیسه های(Vesicles) روکش دار
توزیع شده اند. آنها همچنین در غشای پلاسما انواع سلول تخصصی از جمله استئوکلاست
ها و همچنین سلولهای اپی تلیال در کلیه،مجرای تناسلی مرد،اجسام مویی یا مژگانی
چشمی وجود دارند.PH اسیدی تولید شده بوسیله V-ATpase برای فرایندهای مختلف سلولی از
جمله تفکیک گیرنده و لیگاند آندوزومی،غلظت هورمون،تجزیه لیزوزوم و ترشح اسید مورد
نیاز است. این یافته ها نشان داد که V-ATpase فرایندهای فیزیولوژیکی دارای مرتبه بالاتر را در ارگانیسم های
چندسلولی،علاوه بر نقش سلولی اولیه تقویت می کند.اخیراً،V-ATpase به عنوان یک
مؤلفه ضروری فرایندهای انتقال سیگنال از جمله wnt(مکان یکپارچه
مرتبط به فاقد شاخه)،NoTch و مسیرهای سیگنالینک mTOR، نشان داده شده است. این سیستم های سیگنالینگ در ارتباط با یکدیگر
واکنش تمایزی یا تکثیر سلول را به شبیه سازی های بیرون سلولی تنظیم می کنند. این
بررسی بر توسعه درک ما درباره نقش V-ATpase در سیستم های انتقال سیگنال که موجودات زنده را قادر می سازند تا
ساختار متعدد سلولی را بدست آورند،تمرکز می کند. 2 – ساختار V-ATpase
V-ATpase یک آنزیم دارای چند زیر واحد بسیار حفاظتی است که انرژی حاصل از
هیدرولیز ATP را برای انتقال پروتون ها در سراسر غشاها
مورد استفاده قرار می دهد. شامل دو محدوده اصلی عملکردی V1 و V0 می باشد(شکل 1). اولی دارای هشت
زیرواحد مختلف(A,B,C,D,E,F,G,H) و شامل سه مکان کاتالیزوری برای
هیدرولیز ATP تشکیل شده بوسیله زیرواحدهای A و B می باشد.
محدوده اتصال V0 مسئول انتقال پروتون در سراسر غشاها است. در مخمر شامل بیش از شش زیر واحد e,d,e و پروتئولیپیدهای C می باشد. پروتئولیپیدها بسیار
محافظت شده و دارای چهار پروتئین تراغشایی هستند که در حفره ارگانل دارای termini می باشند و ساختار حلقه مانند را تشکیل می دهند. شش زیر واحد
پروتئولیپیدها در حلقه مبتنی بر آنالیزهای میکروسکوپی شیمیایی و کریوالکترون
کمپلکس کیسه ای(حفره ای) حفاظت شده گاوی و آنالیز میکروسکوپ الکترونی حلقه Nephrops norvegicusc می باشند. چندین پروتئین کمکی با V-ATpase مرتبط و برای یکپارچه شدن کمپلکس
پمپ ضروری هستند. یکی از این پروتئینها،پروتئین AC 8-9 است که شکل ناقصی از گیرنده پرو
رنین رمزگذاری شده بوسیله ATP6ap2 می باشد. بخش ترمینال کربوکسی
ناقص M8/9 مشخص کرد که با V-ATPase مرتبط است و مطالعه ژنتیکی اخیر
درباره AtP6ap2 موش نشان داد که پروتئین دارای طول کامل برای یکپارچگی کمپلکس
ضروری است. M8/9 احتمالاً بوسیله شکافت فیورین(furin) که غیرعملکردی می باشد،تولید شده است.
